レーザ顕微鏡は、光源にレーザを使用し対象物を測定する光学顕微鏡の一種です。
レーザ顕微鏡は、その用途や検出構成によって、さまざまな種類や装置構成が存在し、その内部構成や原理は異なります。
ここでは、一般的なレーザ共焦点顕微鏡を例に、その原理を紹介します。
レーザ光の走査方法や光学系の構成は顕微鏡ごとに異なりますが、レーザ共焦点顕微鏡の特徴として、集光点に配置されるピンホールがあげられます。ピンホールで測定に不要な光を取り除くことで、測定対象の焦点面に非常に近い蛍光のみが検出できます。この構造により、光学的解像度や深さ方向の分解能に優れた測定を実現しています。
浜松ホトニクスでは、レーザ顕微鏡を構成する上で欠かせないキーデバイスを多数製造・開発しています。
取得画像の質に直結するレーザ顕微鏡の心臓部です。より高画質な顕微鏡画像を取得するため、光検出器の中でも感度やノイズ特性に優れた光電子増倍管(PMT)や、MPPC(Si-PM)が多く採用されています。
走査用ミラーとして従来から広く使われてきたガルバノミラーと比べ、小型、低コスト、低消費電力など多くメリットを持つMEMSミラーを製造・開発しています。また、単一デバイスで2次元走査が可能で、装置の小型化や高速化に貢献します。
レーザ顕微鏡用の検出器には、感度とノイズの特性を高いレベルで両立することが求められます。レーザ顕微鏡の測定原理から、検出器へ入射する光量が限られるためです。
例1 レーザ共焦点顕微鏡:検出器への入射光がピンホールによって減光される。
例2 多光子顕微鏡:多光子吸収現象自体の発生確率が非常に低く、光量が限られる。
検出器には多数の種類や機能が存在するため、顕微鏡仕様に適した選定が必要です。下記の画像は種類の異なるPMTを用いて同じサンプル(マウス細胞)を観察したものです。
マルチアルカリ光電面PMTを用いた場合と比較し、結晶光電面のGaAsP-PMTを用いた場合は、よりハッキリとしたコントラストの高い画像が取得できています。このように、一般的には高感度な検出器であるPMTを用いた場合でも、その種類によって取得できる画像に大きな差が生まれます。

浜松ホトニクスは幅広い検出器ラインアップにて、光量、測定波長、サンプルの厚さ、スキャン速度など、多くの基準から最適な検出器を提供します。その中でも、高感度な光電子増倍管はレーザ顕微鏡の画質向上に大きく貢献する性能を有しています。
光電子増倍管に入射した光を電子に変換するパーツ(光電面)にGaAsPやGaAsなどの結晶材料を用いることで、通常のアルカリ材料の光電面を用いたタイプに比べて、高い量子効率を実現しています。
感度波長範囲は、光電面の種類によって定義が異なります。詳細は、カタログのP.04を参照してください。
蛍光分子に光子を2個ほぼ同時に吸収させて励起し、可視・紫外域の蛍光を観測する手法です。
一般の蛍光計測における励起光は蛍光波長よりも短波長ですが、多光子顕微鏡では蛍光波長よりも長波長の近赤外領域の光を用います。近赤外光は可視光よりも透過しやすく、測定対象深くの情報を入手でき、対象内部での散乱や背景ノイズの影響も軽減できます。また、光の持つエネルギーが可視光や紫外光と比較して小さいため、細胞に対するダメージも抑えることができます。

高感度の光電子増倍管(H15460-40相当品)によって、高S/N比での信号検出を実現しています。また、広い受光面積も有しているため、多光子顕微鏡の特長である深部の観察が、広視野で行えるようになりました。

対象物の蛍光の減衰時間(蛍光寿命)を測定する手法です。
蛍光分子がそれぞれ持っている固有の蛍光寿命を時間分解して計測することで、従来の蛍光測定よりも多くの情報を引き出すことが可能です。わずかな時間で起こる小さな蛍光量の変化を計測する必要があるため、検出器には高速応答特性と同時に高い感度が求められます。

主に生体試料の測定に用いられ、光によって刺激を与え、その反応をリアルタイムで観察する手法です。
励起用と刺激用の複数のレーザを使用する光刺激測定では、計測対象光を必要なタイミングで検出する光学設計を組む必要があります。
ゲート機能付き光電子増倍管モジュールは、ゲート動作を電気的に制御可能なため、光検出のタイミングをコントロールでき、光刺激応用向けの顕微鏡搭載デバイスとして最適です。
また、光学系にLCOS-SLMの波面成形技術を用いてビームを分岐することで、1台のレーザで複数箇所へ同時に光刺激が可能となります。

測定対象となる蛍光たんぱく質の蛍光波長の長波長化に伴い、近年注目度が高まっています。可視領域から近赤外領域の広い波長範囲の情報を取得することで、従来は見られなかった現象の観察が期待できます。
検出器においても、広い領域をカバーする感度波長範囲が必要となるため、近赤外領域にも高い感度を持つ光検出器が有効になります。
