多色蛍光イメージングでは、複数の生体情報を同時に取得するために、複数の蛍光プローブを用いた観察が不可欠です。このような観察において、最適な励起波長の選択は実験結果を大きく左右します。
イメージングに使用される蛍光プローブにはそれぞれ固有の最適波長があり(例:GFPは488 nm、mCherryは561 nm付近)、従来の励起光源では利用可能な波長が限られているため、最適波長を選択できず、励起効率の低下やクロストークに悩まされるケースが少なくありませんでした。
一方で、当社のSuperKシリーズは、独自のフォトニック結晶ファイバ技術を用いた、非線形光学効果により、可視域から近赤外にわたる広帯域の連続スペクトルを有しており、専用の波長フィルタアクセサリを接続することで、目的に応じた波長帯を柔軟に取り出すことが可能です。これにより、励起効率を最大化し、クロストークや試料の損傷を最小限に抑えたイメージングが可能となります。また、従来の励起光源では対応が難しかった新規プローブの導入や、多様な解析手法への展開も容易になります。
さらに、波長フィルタアクセサリの一つである、音響光学可変フィルター(AOTF)をベースとした「SuperK SELECT」を接続することで、最大8波長の同時出力に対応可能です。

ライトシート顕微鏡において、ビーム品質は空間分解能を左右します。ライトシートの厚みは、励起光のビーム品質(M²)と光学系のNAに依存するため、ビーム品質が低いとシートが厚くなり、空間分解能が低下してしまいます。
