私たちが一般的に取り扱う光学顕微鏡は、レンズやフィルタなどの光学素子、それらを保持するための機構やステージなどで構成されており、これらを手動あるいは電動で制御することで観察ができるような仕組みになっています。最近の光学顕微鏡は、レーザの輝点を電磁駆動の鏡などで走査して画像を作る共焦点顕微鏡や、レンズ設計を工夫して手動で回転できる補正環を備えた収差補正対物レンズ、ドーナツ状のレーザ光を用いた超解像顕微鏡の一つである誘導放出抑制顕微鏡法(Stimulated emission depletion microscopy; STED)の発明など、より高機能化・複雑化する傾向にあります。そこで当社は、空間光位相変調器(LCOS-SLM)を顕微鏡光学系に導入して波面制御を行うことで、光ビームの方向や光強度分布を自由に変えて、光学顕微鏡をデジタル制御するための手法を検討しています。
中でも、光学系の点光源に対する応答特性を示す点像分布関数(Point spread function; PSF)の、理論計算およびLCOS-SLMによる動的な制御に注目して研究しています。これを実現するためには、レンズ設計をはじめとした高精度な光技術と位相制御の双方が必要になりますが、当社は技術開発を進めて顕微鏡に応用することで、これまでの顕微鏡では得られなかった多点・収差補正・多焦点・被写界深度拡張などの個別あるいは同時実現に成功しています。特に収差補正によって、これまで観察が難しかった生体深部の鮮明な画像が得られるようになっています。最近では、マウス脳機能計測に適用すべくさまざまなPSFを検討しており、世界各国の研究者と共同で研究を進めています。
LCOS-SLMは任意の位相分布を高精度に表現できるため、さまざまな特殊光ビームを生成することができます。この特長を生かし、当社は、LCOS-SLMを用いた光渦など高次モード光の生成・計測・応用の研究を行っています。光渦は、伝搬軸を中心に位相がらせん状に分布しており(下記図:左)、そのため螺旋波面の中心(これを位相特異点と呼ぶ)は強度がゼロとなりドーナツ型のビームパターンが得られます(下記図:右)。さらにドーナツ型強度分布の軌道上に角運動量が存在し、μm~nmサイズの微粒子や分子を光の持つ力で回転させることができます。これらの特長から近年ノーベル賞でも話題になった光マニピュレーションや超解像顕微計測にも利用され、光と物質の相互作用による新しい計測ツールとして注目されています。当社でも光ピンセットの研究に取り組んでおり、高精度な光渦の生成技術を適応し安定したトルクを供給する光渦ピンセットの実現に成功しています。また、発生した光渦を評価するための手法として、波面センサを用いた計測技術も研究しています。光渦ビームには光強度がゼロ、位相が特定できない位相特異点が存在します。特に高次の光渦は光強度がほとんどゼロの領域が大きくなり、特異点の特定が難しいという問題があります。これらの問題をクリアして、特異点を簡単かつ高精度に検出する手法を提案し、実験的な検証を行いました。高次光渦ビームが従来のガウスビームにより散乱媒質を浸透する能力が高いと報告されており、本計測技術は位相特異点の分布とそのダイナミクスの究明を目指して生体深部への光計測の応用が期待されます。
