FRETとはFörster Resonance Energy Transferの略で、2つのタンパク質が近接して存在する場合に、励起状態のドナー分子からもう一方のアクセプター分子へエネルギーが無放射遷移する現象のことです。ライフサイエンス分野においては、一般的に蛍光分子を使用することが多いため、蛍光共鳴エネルギー移動(Fluorescence Resonance Energy Transfer)と呼ばれることもあります。
FRETの効率は、分子同士の距離や角度、スペクトルの重なりの大きさによって左右されます。FRETが起こる分子間の距離において、エネルギー移動効率が50 %でのドナーとアクセプターの距離のことをフェルスター距離と言います。
FRETの測定を行うには、例えば観察したいタンパク質A、タンパク質Bに対して、一方をドナー、もう一方をアクセプターとする蛍光色素を結合し、ドナーの蛍光もしくはアクセプターの蛍光を測定します。ドナーの蛍光を測定する場合、ドナーがアクセプターに近づくと蛍光輝度が減少するため、その減少度合いを測定します。アクセプターの蛍光を測定する場合は、ドナーがアクセプターに近づくとアクセプターの蛍光輝度が増加するため、その増加度合いを測定します。
このように蛍光輝度の変化を観察することでタンパク質間の距離や構造の変化などの相互作用を観察することができます。
